Laminin and Cell Therapy for Muscular Dystrophies

We propose to decipher the organisation of the laminin matrix, its relationship with myogenic precursors proliferation and differentiation, and its role in normal and dystrophic muscle, including on gene expression in vivo in xenografts. We will investigate the presence of laminin isoforms and receptors in normal and dystrophic cultured human muscle cells, as well as in muscle biopsies. We will define the dynamic evolution of laminin isoforms and their possible deregulation in dystrophic conditions by transcriptomic analyses. We will then alter the expression of laminin isoforms and laminin receptors in vitro using siRNAs, in order to improve engraftment of muscle cells for cell therapy and identify new targets for other therapeutic approaches.
